Definition of Topoisomerase I

The topoisomerases I are referred to such a group of enzymes that serves the role of cutting one of the strands of the DNA that exists in the form of double-helical structure. Moreover, they have the ability to relax the strand by removing the coiling and also to reanneal the strands. This class of enzymes is further subdivided into type IA and type IB.

Both these types are structurally as well as mechanistically different from each other. The type IA tends to relax the negative supercoiling of the DNA in the case of prokaryotes. On the other hand, topo IB is responsible for introducing positive supercoils, for separating the genetic material of the daughter cells after replication, and for altering the linking number by the multiple of one.
